The preparation catalysed formation of gamma-glutamyl hydroxamate from L-glutamate, ATP and Mg2+ or Mn2+ with concomitant hydrolysis of ATP to ADP + Pi [1].
We show that the uncoupling of the hydrolysis of gamma-glutamyl hydroxamate and nascent NH(2)OH production from N(4)-hydroxy-CTP formation is more pronounced with the L109A enzyme, relative to the wild-type CTP synthase [2].
The assay monitors GT activity of glutamine synthetase by following the absorbance of the product gamma-glutamyl hydroxamate at 540 nm [3].
Glutamine synthetase activity in muscle was significantly higher in the 14-day exposed group (4.32 mumol gamma-glutamyl hydroxamate formed.g protein-1.min-1) in comparison to normal (1.53 mumol gamma-glutamyl hydroxamate formed.g protein-1.min-1); this parameter had decreased by 40% following 21 days of exposure [4].
