Summary

This is a key enzyme in the N-glycan pathway .

When a N-glycan sugar is attached to a newly synthetized protein by the OST complex, it releases a PP-Dol residue which is de-posphorilated by DOLPP1.

The P-dol molecule obtained can be recycled in the N-glycan biosynthesis pathway, creating a circular process.

Required for efficient N-glycosylation. Necessary for maintaining optimal levels of dolichol-linked oligosaccharides. Hydrolyzes dolichyl pyrophosphate at a very high rate and dolichyl monophosphate at a much lower rate. Does not act on phosphatidate

Subcellular fractionation and immunofluorescence studies localize the enzyme encoded by DOLPP1 to the endoplasmic reticulum of COS cells [1].
The results of protease sensitivity studies with microsomal vesicles from the lpp1Delta/dpp1Delta yeast mutant expressing DOLPP1 are consistent with Dolpp1p having a luminally oriented active site [1].
