Disease relevance of Isoptocarpine

In a previous study, we suggested a dissociation between spontaneous recurrent epileptic seizures (SRS) and hippocampal supragranular mossy fiber sprouting (MFS) in the pilocarpine model of epilepsy (PILO) [1].
Neither L-NAME nor 7-NI affected the convulsions induced by PIL (clonic seizures) or PTZ (clonic and tonic seizures), having no effect on their CD50 values [2].

High impact information on Isoptocarpine

One possible explanation, would be that SRS in the PILO model do not originate in the hippocampus and thus would not depend on MFS [1].
As compared with fellow eyes, the exfoliative eyes showed poorer visual acuity (p < 0.05) and more frequent occurrence of subcapsular cataract (2% vs 8%) [3].

Gene context of Isoptocarpine

We demonstrated that CYP2A6 genotype is a contributor to pilocarpine pharmacokinetics, although the impact of the CYP2A6 polymorphism may be pharmacologically and toxicologically tolerable [4].
