The results presented herein confirm the mutagenicity in Salmonella of 2-nitropropane and the relative inactivity of 1-nitropropane [1].

To investigate whether this is due to endpoint specificity or organospecificity of 2-NP, studies were carried out in the rat in which micronucleus induction (bone marrow and liver) and unscheduled DNA synthesis (UDS) induction (liver) were measured after oral treatment with either nitropropane isomer [2].
The hepatocyte micronucleus assay was shown to distinguish between carcinogenic/non-carcinogenic isomers, such as 2- and 4-acetylaminofluorene (AAF) and 2- and 1-nitropropane (NP) [3].
These data indicate a positive association between increased cell proliferation and hepatocarcinogenesis induced by these two nitropropane isomers [4].
