Disease relevance of ackA

  • Our laboratory previously studied the interaction between nuo and the acetate-producing pathway encoded by ackA-pta in Escherichia coli [1].
  • In Bacillus subtilis, the products of the pta and ackA genes, phosphotransacetylase and acetate kinase, play a crucial role in the production of acetate, one of the most abundant by-products of carbon metabolism in this gram-positive bacterium [2].

High impact information on ackA

  • Expression of the pta gene was increased in the presence of glucose, as has been reported for ackA [2].
  • During the search for unknown factors involved in motility, we have found that expression of the flagellar master operon flhDC is affected by mutations of the pta and ackA genes, encoding phosphotransacetylase and acetate kinase, respectively (S. Shin, J. Sheen, and C. Park, Korean J. Microbiol. 31:504-511, 1993) [3].
  • The ackA-pta mutant has a pfl::lacZ expression level much higher than that of the wild-type strain, and cultures also exhibit the highest ethanol production [1].
  • Conditional ackA silencing reduced carbon flux to acetate and increased heterologous gene expression [4].
  • PTasRNAs targeted against the ackA gene within the acetate kinase-phosphotransacetylase operon (ackA-pta) triggered target mRNA decay and a 78% reduction in AckA activity with high genetic penetrance [4].

Associations of ackA with chemical compounds

  • Although these two enzymes are part of the same pathway, only mutants with inactivated ackA did not grow in the presence of glucose [2].
